I want to show the multiple online users in the map from their respective locations and to have this functionality, if this is already available please help me what are the settings needs to be applied, can anyone here help me out.

Have you read the User Guide and followed the tutorials? The latter goes into a lot of detail on how to do this.

The user guide can be found on the "Downloads" page for the Map Module (as with all of the Module Projects I believe). In this case, the downloads page can be found here: http://www.dotnetnuke.com/Development/Forge/ModuleMap/Downloads/tabid/968/Default.aspx
